PEG-MGF, short for PEGylated Mechano Growth Factor, represents an advanced version of MGF that significantly surpasses the performance of natural MGF. MGF, a splice variant of the IGF gene, enhances the count of stem cells in muscle tissue and facilitates the fusion and maturation of muscle fibers, a crucial process for the development of adult muscle. Unlike natural MGF, which is synthesized locally and remains within the tissue, synthetic MGF is water-based and, when injected intramuscularly, enters the bloodstream. However, MGF maintains its stability in the bloodstream for only a brief period.
 
PEGylation refers to the process of attaching a Polyethylene glycol (PEG) molecule to another larger entity, in this case, MGF. This PEG coating serves as protection, theoretically enabling the MGF to circulate in the bloodstream without degradation.
 
MGF primarily has localized effects in skeletal muscle and requires modification to travel throughout the body. The challenge with synthetic MGF lies in its intramuscular introduction and water-soluble nature, which leads to its rapid entry into the bloodstream where it remains stable for just a few minutes. In contrast, biologically synthesized MGF is produced locally and does not enter the bloodstream, and due to its short-acting nature, stability is not a concern. By PEGylating MGF, it becomes nearly as effective as natural MGF when administered intramuscularly. This is achieved by partially enveloping the peptide with a PEG structure, which can bond to a protein molecule. The PEG groups safeguard the peptide without fully encasing it, allowing the peptide’s active sites to remain available for their biological activity. In this context, the PEG shell acts as a negatively charged barrier against positively charged substances that could impact the protein.
 
Neurological studies have demonstrated that the use of PEGylated MGF yields a longer-lasting and more stable version of the MGF peptide in serum/blood.
